Is there a way to give a VX2000 shot movie a 35mm look or just clean up the digital look? I have Premiere and After Effects if they will help.

lighting lighting lighting, or so i hear.

never done it personally but i've heard thats the big key. I'm sure someone with better advice will post here shortly.

Use the progressive scan mode on the camera

On AfterEffects run the CineLook plugin (purchased separetly check out www.digieffects.com)

that movie i'm working on a poster for "king pathetic creep" was shot on an XL-1. I've got some stills pulled from it to clean up (remove scanlines, adjust sizes and things like that) they look great. I'll have to ask before i could post any, but head over to starsnorth.com in the near future to see them. The key is mostly lighting, but things like what sp70mm said will help too.
